Abstract:
The Nrf2 pathway and autophagy are abnormally activated in response to cellular stress in various types of human cancers. We found that the protein levels of Beclin1 were significantly lower in NSCLC tissue specimens than in paired normal tissue, while the expressions of Nrf2 protein were increased. And the reduced expression of Beclin1 protein level was found to be significantly associated with more advanced TNM stage (P = 0.035), lymph node metastasis (P = 0.017) and distant metastasis (P = 0.005) while the increased expression of Nrf2 protein was associated with larger tumor size (P = 0.032), more advanced TNM stage (P = 0.011), lymph node metastasis (P = 0.045) and distant metastasis (P = 0.013). We also found that there was a strong inverse relationship between Beclin1 and Nrf2 expression in the NSCLC tissues(r=-0.260, P=0.005). Univariate analysis revealed that the tumor size, histological grade, TNM stage, Lymph node metastasis, distant metastasis, Beclin1, Nrf2, and Beclin1-/Nrf2+ expression were all significant prognostic indicators. Multivariate analysis showed that distant metastasis, Beclin1, Nrf2, and Beclin1-/Nrf2+ expression were risk factors for poor survival of NSCLC patients. This study indicated that the abnormal expression of Beclin1 and Nrf2 contributed to the progression of NSCLC.
