Aging has been an enomous social and medical problem.Slowing down aging especially the central nervous system aging,which has been the key of reducing diseases associated with aging.EPO as the issue study of neuroprotecting has the anti-aging prospect to the nervous system,presently in domestic and foreign the correlative studies still loss of information.In the earlier period research, we have confirmed rhEPO has the anti-senile function from various aspects,and this anti-aging effect is dosage dependent.This research based on the conclusion,will use the aging model induced by D- galactose, with the supplement of rhEPO, try to study from the antioxidase aspect whether rhEPO plays the anti-aging role through enhancing the anti- oxidases activity and improving the antioxidation capacity.This research will clear about one aspect of the anti-aging mechanisms of rhEPO,it has the vital theoretical significance, it can provide a experimental reference for the futher studies.ObjectiveTo study whether rhEPO plays the anti-aging effect through enhancing the antioxidases activity, improving the antioxidation capacity..Methods1.Establish the aging model induced by D-gal:30 SD male rats(263±10g)divided into 3 groups in random,the normal control,the aging model,the rhEPO intervene,10 rats in each group.0.9% NaCl 2.5ml/kg neckback IH per day for the normal control group,5% D-gal 125mg/kg neckback IH per day for the left 2 groups,6 weeks in all.2.RhEPO intervene:from the fifth week, rhEPO abdominal injected 3000U/kg per day for the rhEPO intervene group,otherwise 0.9% NaCl abdominal injected 1ml/kg per day for the normal control and the aging model groups,2 weeks in all.3.Morris water maze was used to assess the spatial learning and memory ability between different groups:start with the third day of the sixth week,5 days for training in advance, then carry out the localization underway trial and the space exploration trial in the sixth day,at last analyse two indexes:the escape latency and the times of across platform.4.Assay the T-AOC and the SOD of brain tissue:after the Morris water maze test,the rats were decapitated,separate the brain,prepare the homogenization of brain tissue, testing the T-AOC and the activity of SOD using Pan-wavelength micro plate reade.Results1.Compared with normal control group,the escape latency of aging model group prolonged, the times of across platform reducedT-AOC and the SOD activity of brain tissue decreased markedly compared with the normal control group.2.The escape latency of the rhEPO intervene group is shorter than the aging group the cross platform times of the rhEPO intervene group is more than the aging model group the T-AOC and the SOD activity of brain tissue are significantly higher than the aging model group.Conclusion1.The aging model induced by D-gal is easy and reliable.2.RhEPO has the potential anti-aging effect on the nervous system.3.RhEPO may play the anti-aging effect through enhancing the antioxidant enzyme activity and improving the antioxidant capacity to reduce the radicals, and the generation products of oxidative stress.
